Overview
SS-31 (Elamipretide, MTP-131, Bendavia) is a synthetic tetrapeptide developed by researchers Hazel Szeto and Peter Schiller as a cell-permeable, mitochondria-targeted peptide. Its sequence — D-Arg-Dmt-Lys-Phe-NH₂ — includes dimethyltyrosine (Dmt), which confers antioxidant activity and mitochondrial inner membrane targeting via electrostatic interactions with cardiolipin.
Cardiolipin is a unique phospholipid exclusive to the mitochondrial inner membrane where it serves as a scaffold for respiratory chain supercomplexes. Age-related cardiolipin oxidation and redistribution leads to mitochondrial dysfunction. SS-31's ability to selectively concentrate at the inner mitochondrial membrane and interact with cardiolipin makes it a powerful tool in research on mitochondrial cristae architecture, respiratory chain efficiency, and oxidative stress.
